Effects of Peripartal Sertraline on Maternal Calcium Metabolism and Maternal Bone Health

چکیده

Abstract Lactation taxes mothers’ bone health, causing them to lose up 10% of their mass for milk calcium. Selective serotonin reuptake inhibitor antidepressants (SSRIs) are commonly used during pregnancy and lactation but independently decrease mass. We previously demonstrated that both maternal offspring densities were compromised by treatment with the SSRI fluoxetine throughout lactation. In this study, we examined whether sertraline similarly affect bone. Female C57BL/6 dams (n=23/group) treated (10 mg/kg/d) or a vehicle (DMSO) 41 d (E0-lactation(L) 21). Pups euthanized at weaning (L21). Dams peak (L10, n=4/group), (L21, n=9/group) 3 mo post-weaning (n=10/group) analysis. Sertraline did not dams’ weight gain impact production/consumption, measured weigh-suckle-weigh. Unlike fluoxetine, which increases circulating calcium, reduced serum calcium concentration L21 compared control (p<0.05). Mammary genes altered L10, observed decreased mammary expression transporters Orai1, Spca1, Spca2, Serca2 (p<0.05), tendency Pthlh (a calcium-regulating peptide; p=0.08). intestinal duodenum, Calb1 cytosolic trafficking protein) was increased L10 Trpv6, Pmca1 Spca2 in either tissue, although gland 3.5 times duodenum. Previously, reported markers formation (P1NP) resorption (CTX) unchanged sertraline-treated L21. This is unlike P1NP without altering CTX. Furthermore, control, alter (e.g. Trap, Rank/RankL, Alp, Runx2) femoral post-weaning. Interestingly, utero exposure litter size (5.4 vs 6.8 pups/dam, p<0.01) pup mortality (20% 5% dead pups/litter, first 24 hours postpartum controls, length gestation live birth rate. Exposure surviving offspring, nor it pups weaning, suggesting an adequate supply from dam. These data suggest that, high-dose low-dose peripartal has milder on may reduce could induce failure thrive offspring.

The Effects of Hypothyroidism on Maternal Health

Background and Objective: Hypothyroidism leads to an increased risk of overweight and obesity. This study examined the effect of weight gain caused by hypothyroidism on maternal health (hypertension, gestational diabetes, and miscarriage) for the first time. Materials and Methods: This case-control study consisted of 123 pregnant women with clinical hypothyroidism and 242 non-hypothyroid pregn...

Effect of maternal variable stress on oxidative status and glucose metabolism in pubertal male rats

Introduction: Metabolic disorders are affected by negative stress experiences in the early stages of life. Accordingly, in this study, the effects of stress during pregnancy on oxidative status and glucose homeostasis in pubertal male Wistar rats were investigated. Materials and Methods: After pregnancy, female rats (200±30 g) were divided into 2 groups (6 rats per group) of stress and non-stre...

The Maternal Brain: An Organ with Peripartal Plasticity

The time of pregnancy, birth, and lactation, is characterized by numerous specific alterations in several systems of the maternal body. Peripartum-associated changes in physiology and behavior, as well as their underlying molecular mechanisms, have been the focus of research since decades, but are still far from being entirely understood. Also, there is growing evidence that pregnancy and lacta...

The impact of maternal obesity on maternal and fetal health.

The increasing rate of maternal obesity provides a major challenge to obstetric practice. Maternal obesity can result in negative outcomes for both women and fetuses. The maternal risks during pregnancy include gestational diabetes and preeclampsia. The fetus is at risk for stillbirth and congenital anomalies. Obesity in pregnancy can also affect health later in life for both mother and child. ...
